From a 2007 presentation at Georgia Tech:

MIKE CHAPMAN: We animate everything at 12 frames a second—
MATT CHAPMAN: —'cause that's what you did in 1999.
MIKE: And that's how we learned to animate. We've rarely done a few things at 18 or 24 frames a second, and of course it takes us way longer, so we stick with 12.
MATT: We're glad that the things were the way they were in '99. It definitely makes it much quicker—we're trying to make a three-to-five-minute cartoon pretty much every week, and animating it at 12 frames per second definitely makes that possible.

Most Homestar Runner toons are animated to run at 12 frames per second (FPS). However, some scenes are animated at a higher frame rate for various reasons.
